Therapeutic efficacy of a conjugate vaccine containing a peptide mimotope of cryptococcal capsular polysaccharide glucuronoxylomannan.
|
| description: |
The epitope was selected from a random peptide phage display library by screening with a glucuronoxylomannan (GXM)-specific, protective human mAb (2E9, IgM/lambda), described in the reference Fleuridor et al, J Immunol. 2001 166(2):1087 [PMID:11145689].
Survival was determined and serum samples were collected at a various post-vaccination time points. Chronically or acutely infected mice had longer survival than controls. In the chronic model also lower serum GXM levels at early post-vaccination times was recorded, but not at later points. A similar trend was also observed when peptide was conjugated to the diphtheria toxin (DT). The effects of vaccination on the cytokine production was attributed to the carrier protein conjugate.
