Truncation of the C-terminal 9 residues residues of Der f 2 significantly reduced binding to patient serum IgE. Truncation of the C-terminal 6 or 7 residues also reduced binding. Truncation constructs of Der f 2 with the C-terminal residues (121-129) deleted showed significantly reduced inhibition of patient serum IgE binding to Der f 2. Single alanine substitution of residues in this region did not dramatically reduce IgE binding activity of the mutant Der f 2. The authors conclude that region 121-129 contains part of an IgE-binding epitope. Disruption of the disulfide bond between Cys8-Cys119 may contribute to the reduced binding of the truncation mutants.
