This conserved sequence is present in 972 out of 1088 subtype H5 hemagglutinin sequences analyzed, while none of the 599 H1, 1749 H3, 204 H6, 238 H7 and 184 H9 subtypes hemagglutinin sequences analyzed contained the epitope.
The immunochemistry studies in MCDK cells experimentally infected with H5N1 strains, in formalin-fixed tissues from H5N1 experimentally infected chicken and mouse, and in formalin-fixed tissues from H5N1 naturally infected birds and humans showed specific staining with the epitope-specific mAb. The following H5N1 strains are stated to be detected: human strains Indonesia/CDC594/2006, Indonesia/TLL001/2006, Indonesia/TLL011/2006, Indonesia/TLL012/2006 and avian strains or attenuated strains: turkey/Turkey/1/05, Vietnam/1203PR, Indonesia/TLL005/2006 and Indonesia/TLL101. In addition, other strains tested gave a total of 23 of the 24 human strains and all of five avian H5 strains being successfully detected by 7H10, while none of the samples infected with non-H5 strains presented any positive staining. The 23 human strains detected by the mAb carried either the identical epitope sequence (21/23), or the similar sequence FFWTILNP (2/23), while in the non-detected strain Indonesia/CDC669/2006, the sequence was found to be FFWAILKP.
