The residues are located in the ED3 domain of the envelope protein. These residues were determined to be critical for the definition of the DENV complex-specific epitope. Other residues (T303, D382, A384, K386, R391) are involved but not critical, and are considered to be in the periphery of the epitope.
Affinity towards the expressed recombinant Envelope domain 3 (ED3) was determined for the epitope specific mAb. The residues involved in the epitope were defined by analyzing the relative affinity towards ED3 point mutants at different surface-accessible residues. The same mAb had been tested in a similar way for the binding to DENV-2 ED3, and the region bound was similar, but the involvement of the individual residues in the binding energetics was not conserved.
