The epitope was determined by use of overlapping 15- and 10-mers covering the Ara h 1 sequence. The peptide was tested for recognition of individual patient's serum. With a pool of donors' sera, the reactivity towards single residue substituted peptides was assayed and showed that residues Y67, D68, P69, V72 and Y73 are required for binding, while L65 and E66 substitution diminished binding strength to less than 50% of the WT.
