Neutralization of inhibitor in patient plasma by the epitope was measured. Inhibition of factor IX activity was determined by an activated partial thromboplastin assay using severe hemophilia B plasma as substrate. The epitope was previously shown to neutralize inhibition by patient plasma only at very high concentrations (10mM). Single amino acid substitutions of the epitope decreased the concentration of the peptide needed. One patients’ plasma was neutralized by the substitution peptide A6D (14.5%; 0.5 mM), A6E (9.5%; 0.5 mM), or A6Y (13.3%; 0.5 mM).
