The epitope specificity of the monoclonal antibody was deduced based on binding to Nipah virus glycoprotein deletion mutants. In further assays, triple alanine scan mutants in the epitopic region none of the triple alanine mutants showed a significant decrease in Mab45 binding. The authors suggest this may indicate that the Mab45 epitope was complex and was not determined by any short primary linear sequence, or that deletion of this region may have caused conformational changes in the protein preventing epitope binding by the monoclonal antibody.
