Liposomes prepared from rabbit RBC membranes, expressing the epitope on its suface and containing encapsulated hen egg ovalbumin (OVA) are tested in ELISA with sera from WT and alpha1,3GT KO mice immunized with porcine kidney membranes. All 6 PKM immunized KO mice displayed high anti-Gal IgG activity. In contrast, PKM immunized WT mice displayed no IgG binding to OVA–liposomes. Since KO and WT mice differ immunologically only in their ability to produce anti-Gal, the lack of IgG binding to OVA–liposomes in WT mouse sera implies that anti-Gal is the only Ab in KO mice that binds to OVA–liposomes and that no other Abs are produced against these liposomes.
