Only 2 out 24 allergic patients sera (8%) showed a significant drop in IgE binding to below 80% of the reactivity of the wild type protein in a dot-blot of mutants R30A, R32A, and K81Amutants R24A, R26A and K75A, and a more minor drop was found for mutant K86A. An ELISA with serum from a selected patient also showed that these mutations did not cause significant reduction in IgE binding, indicating that these residues may not be major epitopes of Bla g 4.
