About 50% of the allergic patients sera showed a significant drop in IgE binding to below 80% of the reactivity of the wild type protein in a dot-blot of single residue mutants where the epitopic residues were changed to alanine. An ELISA with serum from a selected patient also confirmed these findings, indicating that these residues are essential residues for IgE binding in Per a 4.
