The epitope recognized by the human mAb was deduced based on reactivity with GAD65/67 chimeric proteins and single amino acid substitutions replacing GAD65 residues with that found in GAD67. Single amino acid substitutions E517P or E521Q abolished reactivity with M2, indicating these are critical residues.
