Distinct antigenic features of linear epitopes at the N-terminus and C-terminus of 65 kDa glutamic acid decarboxylase (GAD65): implications for autoantigen modification during pathogenesis.
|
| description: |
The epitope sequence is also found in GAD67 (residues 581-594).
The rabbit anti-epitope antiserum selected the peptide from a T7 phage library expressing 9-mer peptide inserts constrained by a difulfide bridge after immunopanning. The peptide had homology to GAD65 residues 582-585. The suggested consensus epitope for the C-pc antibody is GQDL, spanning residues 582-585 of GAD65. Analysis of the immunoselected peptides suggests that the C-pc polyclonal antibodies recognize two distinct sites within the epitope sequence.
