The epitope was deduced through assays with protein constructs mutated and deleted for amino acids corresponding to the epitope.
The epitope specific monoclonal antibody bound the botulinum neurotoxin type A1 and several fragments from botulinum neurotoxin type A1, aa125-200, aa65-200, and 111-200 on a Western blot. Amino acids corresponding to the epitope, aa139-141 were found to be critical for binding by Western blot and competition with proteins that were mutated and deleted for those residues. Residues R145 and S146 also contributed to antibody binding as shown by reduced binding to the glycine substituted peptide.
