This epitope was defined through analysis of mutated glutamate decarboxylase 2 binding to a human monoclonal antibody.
The epitope specific monoclonal antibody bound glutamate decarboxylase 2. Amino acids 524-526, 572-573, and 498-500 were shown to be important for binding by alanine mutagenesis. Binding of gluatmine decarboxylase 2 to GAD1 and GAD6 monoclonal antibodies was also affected by mutating 572-573. This indicates that this mutation has structural effects rather than direct contact with the monoclonal antibody.
