Competitive binding ELISA was used to define antigenic regions. Indirect fluorescent assay was used to demonstrate that replacement of the epitope with the sequence from bunyamwera virus G2 caused a complete loss of binding, indicating it contributes to a discontinuous epitope. Replacement of other regions (29-34 and 50-58) also reduced binding, indicating they also contribute to the discontinuous epitope. G2 protein was expressed in recombinant vaccinia-T7 virus-infected HeLa cells.
