The epitope was deduced.
The epitope was deduced based on reactivity of Bermuda grass pollen-allergic patients' sera with truncated fragments of Cyn d 1. Pooled (n = 10) and individual sera reacted in a dot bot assay with fragment 1-170, but not with fragment 1-120, indicating the region 121-170 contained an IgE epitope. This was confirmed by reduced IgE reactivity (46% reduction) with Cyn d 1 deletion mutant lacking residues 120-170 by ELISA.
