alpha-gal was coupled to sepharose via a linker (Galalpha1-3Galbeta-OCH2CH2CH2NH-sepharose) and used in direct ELISA and pre-adsorption experiments. All donors had alpha-gal serum IgEs, and there is a significant correlation between IgE antibodies to alpha-gal and IgE antibodies to beef. Reactivity to beef, pork, cat and dog extracts was significantly reduced by pre-adsorption of the sera with the epitope, while no effect was seen on the reactivity to non-mammalian extracts.
