Recognition of WT Env protein by the epitope-specific mAb occurred only under non-denaturing conditions, confirming the conformational nature of the epitope. Recognition of recombinant truncated Env including only domains I and II in a yeast-display system was abrogated for mutant proteins with substitutions at residues W101, G104 and G106. Recognition of WT Env was detected also in ELISA.
