The epitope recognized by human mAb M10 was deduced based on reactivity with GAD65/67 chimeric proteins. Swapping residues 242-282 of GAD65 with those from GAD67 abolished reactivity with M10. Single amino acid substitution of E264T in this region abolished reactivity with M10, indicating it is a critical residue.
