The epitope recognized by the human mAbs was deduced based on reactivity with GAD65/67 chimeric proteins. Swapping residues 532-540 of GAD65 with those from GAD67 abolished reactivity with M8 and M9. Single amino acid substitution of R536L or Y540S abolished reactivity with M8 and M9, indicating they are critical residues for these mAbs. Single amino acid substitution H470Q also reduces M8 and M9 reactivity, indicating that other regions affect the epitope.
