The epitope is a mimotope of discontinuous IgE binding epitope on Tri a 14 including residues L34, H35, N36, R39, S40, S42, D43, P70, L77, P78, Y79, and T80.
The epitope sequence was selected from a random peptide phage-displayed library by pooled serum IgE from patients with baker's asthma caused by wheat flour (Tri a 14) and allergy to peach fruit (Pru p 3) and eluted with Tri a 14. Reactivity with patient IgE was confirmed by ELISA. The selected phage sequences were mapped to a consensus sequence on the surface of Tri a 14 corresponding to residues L34, H35, N36, R39, S40, S42, D43, P70, L77, P78, Y79, and T80. Comparing sequences of peptides eluted with Tri a 14 after elution with Pru p 3, the authors deduced a final epitope corresponding to residues H35, N36, R39, S40, S42, D43, G74, V75, L77, P78, Y79, and T80.
