The epitope-specific mAb was tested for binding to purified HPV 16 virions containing a wild-type and mutated epitope in the L2 protein. The mAb showed positive binding to the wild-type epitope. Single alanine substitutions were made to the epitope. Single substitutions K4A, C6A and C12A eliminated mAb binding and P13A greatly reduced mAb recognition.
