The epitope of HBV non-particulate e-antigen was N-terminally extended by an acetyl-thio-acetyl group.
The minimal epitope recognized by mAb anti-HBe4 was determined by direct binding or by competition ELISA with C-terminally truncated peptides. The 12-mer epitope was the minimal truncation of the 17-mer peptide (LEDPASRDLVVNYVNTN) that inhibited binding. The 15-mer (LEDPASRDLVVNYVN) was required for direct binding. The peptides were N-terminally extended with an acetyl-thio-acetyl group, as no signal was detected with unmodified peptides.
