The epitope residues were calculated from the structure [PDB: 1ZA3] as antigen residues interacting with Fab at atomic distance of 4 Å
. Mapping of residue numbers from PDB to external identifier was done as provided in the PDB.
The epitope recognized by Fab-YSd1 on hDR5 extracellular domain was determined by X-ray diffraction and solved by molecular replacement analysis. Despite the modest resolution of the structure, the electron density maps are of high quality and allow for accurate determination of backbone and side chain positions. There are two Fab-antigen complexes in the asymmetric unit. Fab binds to the N-terminal portion of hDR5 and occludes the smaller of two binding sites for the natural hDR5 ligand Apo2L/TRAIL.
