The epitope residues recognized by mAb 1H7 were deduced based on reactivity with human TSHR mutated at one or more residues or chimeras of receptors for TSHR and LH/CG. The mAb recognizes region 1-281 of hTSHR, as well as TSHR from mouse, rat, cat, dog, and sheep. Mutation of single residues T56, K58, R80, Y82, or R109 abolished binding, indicating these are critical residues. In addition, the triple mutation of F130, G132, and F134 reduced binding, indicating these residues contribute to the antibody binding site. MAb 1H7 inhibits the binding of bovine TSH to the TSH receptor in a dose-dependent manner.
