Human monoclonal Fab fragments (rhFabs) specific for the immunodominant region B (IDR-B) bound to wild-type TPO and selected point mutants by ELISA. Single amino acid mutant K627A abolished binding for rhFabs 126TP5, 126TP14, and 131TP7, demonstrating this is a key residue in IDR-B. Point mutant Y772A also showed reduced binding, but the authors suggest that this mutation may effect the overall tertiary structure of TPO.
