Human monoclonal Fab fragments (rhFabs) specific for the immunodominant region A (IDR-A) bound to wild-type TPO and selected point mutants by FACS. Triple mutant R225A/R646A/D707 reduced binding approximately 55-fold. Double mutants R225A/R646A, R225A/D707, or R646A/D707 reduced binding approximately 3- to 6-fold. The results were confirmed by ELISA, which showed greater sensitivity to the mutations with single mutations R225A, D707N and R646A showing significantly reduced binding The authors conclude that residues R225, R646, and D707 contribute to the IDR-A epitope.
