MAb IRI-SAb2, which recognizes the epitopic residues, increased total T4 thyroxine levels and decreased TSH levels in recipient mice compared to controls. Levels were altered as early as 8 h after injection and remained altered at 4 days. The T4 level returned to normal at 7 days, consistent with the serum half-life of of mouse IgG2a. Thyroid morphology showed hypertrophic cells in the follicular epithelial layers and necrotic thyrocytes in the follicular lumina. CD45+ immune cells and Mac-1+ macrophages were observed between the thyrocytes and inside the colloid. The mAb induced hyperthyroidism.
