Rabbit anti-epitope antibody (anti-P14) competed sera from patients with Hashimoto's thyroiditis from binding to human TPO in a competitive ELISA. The anti-P14 antibody is used to define immunodominant region B (IDR-B). The percentage of antibodies to IDR-B was significantly lower in sera of the HT group (50 +/- 15%) compared to the healthy control group (66 +/- 22%). A slight decrease in IDR-B reactivity with increasing TPO antibody level was observed. IDR-B reactivity was not dependent on patient age.
