The ability of MAb 1A1D-2 to neutralize DENV infectivity was quantified using a plaque reduction neutralization test (PRNT). The PRNT50 concentration with DENV-2 strain NGC was 2.1±0.1 nM. In experiments to assess ED3 occupancy on the virion, the authors found that the majority of virus infectivity was neutralized at relatively high occupancy levels. Mutations at residues K307, K310 and I312 completely abrogated neutralization activity.
