Specific antibody-forming cell responses were measured in mouse spleen cells by using epitope-coupled sheep erythrocytes (SRBC) as indicator cells in the haemolytic plaque assay. Immunization with the combination of FITC-HGG with LPS induced the formation of IgG-plaque forming cells primarily of IgG1 and IgG2b subclasses, while the injection of epitope together with Freund's adjuvants gave a marked increase in IgG antibodies, with a strict preference for IgG1.
