The epitope is a widely used His tag, consisting of 6 consecutive histidine residues.
Epitope specific 3D5 scFv (mut1+2) recognized a mouse scFv fragment (anti-HER2 antibody 4D5) with a His tag of three, four or six histidine residues at the C-terminus. It did not recognize a scFv with six histidine residues followed by a glycine residue. It was concluded that 3D5 recognizes 3 histidine residues and the last residue must be histidine.
