Serum anti-DNP antibody levels were determined by a modified Farr technique using DNP-tritiated thymidine-amino-N-caproic acid. Mice immunized with DNP-KLH and boosted two weeks later with DNP-KLH or DNP-BGG produced DNP-specific antibodies. This was not true of BALB/c mice immunized with DNP-carrier. Adoptive transfer of splenic lymphoid cells from non-immunized mice enhanced this response by way of the allogeneic effect related to graft-versus-host reaction. Route of administration, number of transferred cells and interval of secondary transfer all were found to be pertinent factors in the magnitude of response.
