Serum IgG from MS patients reacted better with the peptide glucosylated (epitope) at position 7 than serum IgG from healthy controls. The unglycosylated peptide or peptide with the following glycosyl moieties at Asn7 did not bind serum IgG or detect an increase in binding in MS patients' sera: galactose, mannose, N-acetylneuraminic acid and ribose. The peptide with a Asn7Gln, but not Asn7Phe or Asn7Gly substitution, and modification with the epitope at that position also bound to MS patients' sera IgG.
