The epitope inhibited cytoxocity of human sera against porcine aortic endothelian cells (PEC-A). The epitope was expressed on the extracellular portion of a cell surface-anchored mucin, P-selectin glycoprotein ligand-1 (PSGL-1) fused to an immunoglobulin Fc portion to form the PSGL1/mIgG2b construct. Absorption of human sera with PSGL1/mIgG2b inhibited complement-dependent cytotoxicity against PEC-A and depleted reactivity with all membrane components of PAC-A cells. The decrease in cytoxicity correlated with the decrease in reactivity to PAC-A cell components. The inhibition was greater with the PSGL1/mIgG2b construct compared to the epitope conjugated to agarose beads or macroporous glass.
