The epitope residues are calculated from the PDB structures as the antigen residues at 4 Å
distance from the antibody.
The epitope of Fab 10E5 on integrin alphaIIbß3 was determined from the crystal structure of the complex. Fab 10E5 binds solely to the alphaIIb cap subdomain and has no effect on its conformation. The peptide LGGAKQRGDV was soaked into crystals of the soluble headpiece of integrin alphaIIbbeta3 (chain A: Integrin alphaIIb chain B: Integrin beta3) and the 10E5 Fab. Also, other structures were obtained by soaking other peptides, 2VDP, 2VDQ, 2VDO, 2VDN, and without peptides, 2VC2, 2VDK, 2VDL, 2VDM. Calculations show that the epitopes were identical in all structures; therefore, only one structure with the best resolution was curated.
