name:
Structural and functional analysis of a C3b-specific antibody that selectively inhibits the alternative pathway of complement.
description:
The epitope residues are calculated from [PDB: 3G6J] at 4 Å
distance. Epitope is located on alpha and beta-chains of C3b. PDB chain B is the C3b alpha-chain, PDB chain A is the beta-chain. Epitope residue on beta-chain in GenBank numbering [GI:119370332] are S570, R573, V576
on alpha-chain (same GI number) are D754, D828, R855, N857, Q858, E859, K861, P890, H918, H919, F920, I921.
The epitope specific S77 Fab bound to complement C3b with high affinity and bound to hydrolyzed C3 (iC3; KD = 1.2 nM), iC3b (KD = 2.0 nM) and C3c (KD = 2.2 nM). S77 did not bind significantly to C3 by ELISA.
