The epitope specific disulfide-free anti-RAS, VH#6(AV) and VL#204(VA) (designated HRAS-Fv SS-free) reverted RAS-dependent transformation of NIH3T3-EJ cells as shown using an achorage independence assay. Expression of disulfide-free anti-RAS VH#6(AV) in NIH3T3-EJ cells inhibited soft agar colony formation. Expression of VH#6(AV) in NIH3T3-EJ cells also inhibited lung tumor development in vivo, assayed 14 days after injection of the cells.
