name:
Combinatorial optimization of a CD4-mimetic miniprotein and cocrystal structures with HIV-1 gp120 envelope glycoprotein.
description:
The epitope residues are calculated from the PDB structure [PDB: 2I5Y] as the antigen residues at 4 Å
distance from the antibody.
The crystal structure of epitope-specific monoclonal antibody bound to GP120 core (HIV YU2 isolate) and the CD4 mimetic peptide was determined. The GP120 core protein had Gly-Ala-Gly tri-peptide substitutions for the V1/V2 and V3 loop regions. The scorpion toxin mimic of CD4 (CD4M47) contained beta-mercaptopropionic acid at position 1, (R)-2-amino-3-(4-phenylcyclohexyl) propanoic acid at position 23 and valinylamine at position 27. There are no direct contacts of 17b Fab with CD4M47.
