Epitope residues are calculated from the structure [PDB: 3MXW] as the antigen residues at 4 Å
distance from the antibody.
The epitope specific Fab ch5E1 bound to Sonic hedgehog (Shh) with an affinity of 6.94 nM, as measured by biolayer interferometry. Addition of Ca2+ improved the affinity to 1.12 nM and Ca2+ plus Zn2+ further improved the affinity to 0.31 nM. The increase in affinity was mainly due to a decrease in the dissociation rate constant.
