The epitope-specific mAb stained cells of Cryptococcus neoformans serotype A, strains CN6, MU-1 and H99. CN6 cells were grown in synthetic medium or isolated from brains of infected mice. In cells grown in synthetic medium, there was considerable variability in binding to cells. Some cells showed a pattern of bright binding to the capsule that surrounded the parent cell; other cells showed a fine punctate binding pattern, but most cells failed to bind the antibody. Binding was bright and uniform in cells isolated from infected brains and in cells grown in FBS. The mAb did not stain capsules surrounding new buds of cells. Binding to parent cells increased in intensity as the cells aged. Staining was lost when cells were de-O-acetylated.
