The epitope footprint of the epitope-reactive GA101 mAb was determined by pepscan of overlapping adjacent 8-mer peptides spanning the sequence of the CD20 large extracellular loop, residues 142-187 (with Cys to Ala substitutions). The footprint of GA101, the parent mouse mAb B-Ly1, and an unrelated type II anti-CD20 mAb H299/B1, started around position Ala167 and extended to Pro178. Single alanine substitutions demonstrated that the core epitope of GA101 was formed by residues 172-178 (PSEKNSP).
