The epitope-reactive GA101 mAb bound to CD20 transfected into Freestyle 293-F cells. Binding was not significantly affected by mutation of residue Asn171, whereas Ala substitution of Asn176 reduced binding. GA101 also bound to CD20 on Z138, Ramos and normal B cells and induced an accumulation of GA101-CD20 complexes at sites of cell-cell contact.
