The epitope footprint of the epitope-specific Rituximab mAb was determined by pepscan of overlapping adjacent 8-mer peptides spanning the sequence of the CD20 large extracellular loop, residues 142-187 (with Cys to Ala substitutions). The footprint of Rituximab, and the unrelated type I anti-CD20 mAbs C2H7 and LT20, started around position Ile162 and extended to Asn176. Single alanine substitutions demonstrated that the core epitope of Rituximab was formed by residues 168-175.
