The antibody used was mutated at residues N52K, N53H and A55T of the light chain, which resulted in increased affinity for the epitope compared to the germline antibody. Binding by variants of the epitope-specific monoclonal antibody was also measured. The sFv measured 2.8 x 10-3 nM-1 and the germline antibody measured 3 x 10-5 nM-1.
