An antibody competition study was performed to determine the binding site of recombinant IgG1 from a patient chronically infected with HCV. Alanine mutagenesis scanning identified the epitope sequence based on the greatest reduction in binding. Residues S424, G523, P525, D535, V538 and N540 were identified as being critical for binding.
