The epitope was designed to mimic discontinuous antigenic site D of FMDV isolate C-S8c1, which encompasses residues from VP1 (Thr193), VP2 (Ser72, Asn74, His79 in the sequence PSQNFGHMHKVVLP), and VP3 (Glu58 in the sequence FLMFENVPY). The epitope D8A, which represents the VP2 and VP3 peptides linked by a polyproline spacer, was disulfide bonded to peptide D8B (CPPPPTG), which mimics the VP1 peptide.
One out of three animals immunized with the D8 epitope disulfide hetetodimer was protected against challenge with FMDV, and a second animal developed only minor lesions at 5 days post challenge.
