The cysteine residue at position 1 was made as the 3-nitro-2-pyridylsulfenyl derivative. The epitope was designed to mimic discontinuous antigenic site D of FMDV isolate C-S8c1, which encompasses residues from VP1 (Thr193), VP2 (Ser72, Asn74, His79 in the sequence PSQNFGHMHKVVLP), and VP3 (Glu58 in the sequence FLMFENVPY).
The binding of epitope-reactive anti-D8 sera to FMDV was inhibited by FMDV site D-specific mAbs 5C4 and 2A12, demonstrating the specificity of the anti-D8 sera.
