The binding of serum IgE from mite-allergic patients to pro-Der p 1 was reduced 20-95% by single amino acid mutations R156A, Y185V or D198A. Pro-Der p 1 inhibited the binding of pooled sera from four donors with the strongest reduction in binding to the mutations. Pro-Der p 1 with mutation R156A, Y185V or D198A were 5- to 10-fold less effective at inhibiting the binding of the serum pool to pro-Der p 1.
