The epitope inhibited the binding of the antiserum to HIV-1 HXB2 gp41. Antiserum from two rabbits also bound directly to the 4E10 epitope peptide. Alanine substitution of Trp, Phe, Asp, Ile or Leu at positions 2, 3, 4, 5 and 9, respectively, significantly reduced avidity of the antisera for the epitope.
